A Functional GM-CSF Receptor on Dendritic Cells Is Required for Efficient Protective Anti-Tumor Immunity

Dendritic cells (DC) play a major role during the priming phase of anti-tumor immunization, as they are required for an efficient tumor-associated antigens presentation. At least one dendritic cell-based therapy has already been successfully approved by regulators clinical application in prostate cancer patients. Moreover, DC development is dependent on granulocyte macrophage colony stimulating factor (GM-CSF), cytokine that used potent inducer anti-tumoral immunity. To better understand relation between and GM-CSF immunity, we studied function mice lacking receptor common subunit beta (βc-/-) GM-CSF, IL-3 IL-5 immunized with irradiated tumor cells. Such immunization induces protective, specific wild-type mice, while βc-/- failed to mount immune response. Upon vitro stimulation, from (DCβc-/-) unable undergo full maturation level. In vivo experiments show lack ability prevent growth, contrast DCWT. matured DCWT rescued mice. functional pathway involving signaling through biologically receptor. These findings may contribute new strategies immunotherapies.
